An antibody specific for EFEMP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyEFEMP2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for EFEMP2 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EFEMP2 bound in the initial step. The color development is stopped and the intensity of the color is measured. FBLN4 contains four EGF2 domains and six calcium-binding EGF2 domains. This gene is necessary for elastic fiber formation and connective tissue development. Defects in this gene are cause of an autosomal recessive cutis laxa syndrome.EFEMP1 is likewise expressed in a wide range of adµLt and fetal tissues. In contrast to EFEMP1, however, EFEMP2 was not significantly overexpressed in senescent or quiescent fibroblasts, sµggesting a diversity of function within this EGF-like domain subfamily.
