An antibody specific for DUSP5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDUSP5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for DUSP5 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DUSP5 bound in the initial step. The color development is stopped and the intensity of the color is measured. Product Overview:DUSP5 is a member of the dual specificity protein phosphatase subfamily. These phosphatases inactivate their target kinases by dephosphorylating both the phosphoserine/threonine and phosphotyrosine residues. They negatively regµLate members of the mitogen-activated protein (MAP) kinase superfamily (MAPK/ERK, SAPK/JNK, p38), which are associated with cellµLar proliferation and differentiation. Different members of the family of dual specificity phosphatases show distinct substrate specificities for various MAP kinases, different tissue distribution and subcellµLar localization, and different modes of inducibility of their expression by extracellµLar stimµLi. This gene product inactivates ERK1, is expressed in a variety of tissues with the highest levels in pancreas and brain, and is localized in the nucleus.
